Strange problem when enabling pretty URLs

I have done dozens of sites with Concrete5 but have just come across a strange problem. The site works fine until I enable Pretty URLs, once enabled when you navigate to another page all I get is a repeat of the home page but all messed up. I have attached a screen to show how it displays.
I wonder if it may be to do with the cache as when I look at an image the path is
about-us/who-we-are/files/cache/41850ee6be81970fe29080c73afcc994.jpg
which can't be found. Strange thing is I havent got the cache enabled. Also the home page displays fine even with Pretty URLS enabled
Any clues.

Grrrrr, it was me. Noticed something was missing from define('DIR_REL', '');!!!!

I seem to be having the same problem, and can't seem to figure out how to fix it by your posts.
I just enabled pretty urls on this site: tte20.com, and now every page jumps back to home page.
Any idea how to fix this?

noticed that they are just opening the home page but messed up . Probably something obvious but for the life of me I can't figure it out!!!
